Vacation in the Florida Keys (When such things are again possible)

My wife and I were discussing places in the US that we've always wanted to holiday, and the Keys came up again.

 

What are some must-see places or experiences that shouldn't be missed?  What is the best time of year to go?  Fly into Miami and drive down, or is there a better choice?
